Former England batsman and Hampshire captain Robin Smith made an unbeaten 88 as the visitors cruised past Durham's total of 161 for eight with 5.5 overs and seven wickets in hand.
Shane Warne took two for 19 and also made 29 runs, including the game's only six, hit over long-on in a second-wicket stand of 65 with Smith.
It was announced just before the match that Nick Speak had been relieved of the Durham captaincy. Jon Lewis will lead the side for the rest of the season.
Lewis chose to bat and made the highest score of the Durham innings - an unbeaten 40, anchoring the innings after a slide from 81 for two to 105 for six.
The Hampshire attack kept things tight for most of the Durham innings. Alex Morris was the one weak link, giving away 52 runs from nine overs as Durham started briskly and accelerated briefly at the end.
Alan Mullally quietened things down with a spell of one for 13 in six overs, and apart from Lewis Michael Gough was the only Durham batsman to make a worthwhile contribution, reaching 36 - his best score in the competition.
